Abstract
Osteosarkom, gençlerde görülmesi, sağkalım süresinin düşük olması ve kullanılan antikanser ilaçların yan etki profilinin yüksek olması sebebiyle çoğunlukla tedavisi zor bir hastalıktır. Mevcut kemoterapötiklerin kullanımındaki sınırlamaları aşmak amacıyla bu proje bilinen antineoplastik bir ajan olan cisplatin üzerinden bir ilaç taşıyıcı sistem (İTS) formülasyonunun geliştirilmesi üzerine yapılacaktır. İTS'ler son yıllarda yoğun bir şekilde çalışılan, multidisipliner bir alan olarak, akıllı ilaç tasarımlarında önemli rol oynayan teknolojilerdir. Özellikle kanser çeşitlerine yönelik konvansiyonel ilaçların tedavi başarısını arttırmak için lipozom, misel, polimer, jel, altın nanoparçacık, niozom, katı lipid nanopartikül, dendrimer, nanokapsül gibi çeşitli sistemler geliştirilebilmektedir. Bu çalışmada tasarlanan ilaç taşıyıcı sistem için kopolimerik misel yapısı oluşturacaktır. Polimerde 1,1-di-tert-butil-3-(2-(metakriloyloksi)etil)-butan-1,1,3-trikarboksilat (MAETC) monomeri aracılığıyla hem ilacın biyoyararlanımının arttırması hem de cisplatin ilaç molekülünü taşıması, ikinci monomer olan 3-(trimetilsillil) prop-2-in-1-il polimetakrilat (TMSpMA) monomeri ucuna hedeflendirici peptid molekülü eklenmesi, 2-hidroksi etil metakrilat (HEMA) ile hidrofilik özellik kazandırılması ve neticede kendiliğinden misel oluşturacak bir sistemin geliştirilmesi planlanmaktadır.

Osteosarcoma is a difficult disease to treat due to its occurrence in adolescents, low survival time and high side effect profile of anticancer drugs used. In order to overcome the limitations in the use of existing chemotherapeutics, this project will be carried out on the development of a drug delivery system (DDS) formulation based on cisplatin, a wellknown antineoplastic agent. DDSs, as a multidisciplinary field that has been studied intensively in recent years, are technologies that play an important role in smart drug designs. Various systems such as liposomes, micelles, polymers, gels, gold nanoparticles, niosomes, solid lipid nanoparticles, dendrimers, nanocapsules can be developed to increase the treatment success of conventional drugs, especially for cancer types. The drug designed in this study will create a copolymeric micellar structure for the delivery system. It is planned to increase the bioavailability of the drug and carry the cisplatin drug molecule through the 1,1-di-tert-butyl-3-(2-(methacryloyloxy)ethyl)-buthane-1,1,3-tricarboxylate (MAETC) monomer in the polymer, to add a targeting peptide molecule to the end of the second monomer, 3-(trimethylsillyl) prop-2-in-1-yl polymethacrylate (TMSpMA), to gain hydrophilic properties with 2-hydroxy ethyl methacrylate (HEMA), and ultimately to develop a system that will form micelle with self-assembly.
