    Effects of Nigella sativa on apoptosis and GABA(A) receptor density in cerebral cortical and hippocampal neurons in pentylenetetrazol induced kindling in rats

    We investigated the effects of Nigella sativa on apoptosis and gamma-aminobutyric acid (GABA(A)) receptor density in cerebral cortical and hippocampal neurons in a pentylenetetrazol (PTZ)-induced kindling model in rats. The PTZ kindling model was produced by injecting PTZ in subconvulsive doses to rats on days 1, 3, 5, 8, 10, 12, 15, 17, 19, 22 and 24 of the study into animals of PTZ treated (PTZ) and PTZ + N. sativa treated (PTZ + NS) groups. Clonic and tonic seizures were induced by injecting a convulsive dose of PTZ on day 26 of the study. Rats in the PTZ + NS group were treated also with a 10 mg/kg methanolic extract of N. sativa 2 h before each PTZ injection. Rats in the control group were treated with 4 ml/kg saline. The number of neurons that expressed GABA(A) receptors in the hippocampus and cerebral cortex of rats in the PTZ and PTZ + NS groups increased significantly. There was no significant difference in the number of GABA(A) receptors between the PTZ and PTZ + NS groups. GABA(A) receptor density of the neurons in the cerebral cortex, but not hippocampus, was increased in PTZ group compared to controls. We observed a significant increase in the number of apoptotic neurons in the cerebral cortex of rats of both the PTZ and PTZ + NS groups compared to controls. We observed a significant decrease in the number of the apoptotic neurons in the cerebral cortex of rats in the PTZ + NS group compared to the PTZ group. N. sativa treatment ameliorated the PTZ induced neurodegeneration in the cerebral cortex as reflected by neuronal apoptosis and neuronal GABA(A) receptor frequency.

    Impaired memory by hippocampal oxidative stress in rats exposed to 900 MHz electromagnetic fields is ameliorated by thymoquinone

    The protective effects of thymoquinone on learning and memory abilities and on hippocampal oxidative stress in rats exposed to 900 MHz electromagnetic field radiation at s strength of 6.0 +/- 0.5 V/m for 1 h on each of 15 days was studied. Thymoquinone-treatment (10 mg/kg/day) of radiation-exposed rats resulted in better performance than that of radiation-exposed rats not receiving thymoquinone. Electromagnetic field-exposure led to increased hippocampal superoxide dismutase, glutathione peroxidase, and malondialdehyde levels and to lowered brain-derived neurotrophic factor level; thymoquinone treatment ameliorated all four parameters. Also, in the electromagnetic field-exposed group, N-methyl-D-aspartate receptor expression decreased, and gamma-aminobutyric-acid alpha receptor expression increased, which was reversed by thymoquinone treatment. In conclusion, treatment with the thymoquinone ameliorated the impact of the exposure to EMF and provided a hint on the mechanism involving oxidative stress.

    Effects of Thymoquinone On Erythrocyte Aggregation Kinetics: An In Vitro Study

All rights reserved.Erythrocyte aggregation is associated with a series of physio-pathological processes in body and according to this, current study was designed to examine how the thymoquinone (TQ) and a ggregation kinetics relation changes when the aggregation properties of erythrocyte suspensions are intervened using different approaches, such as changing the suspension environment or erythrocyte surface properties. Blood samples from 40 male Wistar albino rats (three months old weighing 250-300 g) were randomly divided into four groups, as 1) whole blood (control), 2) 1/2 dilution (plasma was diluted with PBS), 3) dextran 500 (1% dextran 500 was dissolved in plasma) and 4) glutaraldehyde. Blood samples were taken from abdominal aorta under anesthesia. The erythrocyte suspensions were incubated at 37°C for one hour in the absence or presence of TQ, with a final concentration of 1 mg/ml. Then, the electrical properties of erythrocyte suspensions were recorded, and the electrical capacitance was monitored using an inductance, capacitance, and resistance meter (LCR meter) at 100 kHz. It was found that TQ incubation decreased the aggregation index in control and dextran groups while increased it in 1/2 dilution and GA groups. It was also found that TQ incubation increased the time constant in whole blood and dextran while decreased it in 1/2 dilution and GA. The TQ can normalize the altered erythrocyte aggregation responses in both ways that is, it decreases the increased aggregation or increases the decreased aggregation. Therefore, it may have very critical role in rouleaux formation-induced circulatory disorders.
