In total, 56 patients with cataracts and 49 healthy controls were enrolled in this case-control study. Serum total thiol (TT), native thiol (NT), and disulfide (DS) concentrations were determined using a novel automated measurement method. Additionally, DS/TT, DS/NT and NT/TT percentage ratios were compared between the groups. In comparison with the control group, serum NT levels and aqueous humor TT and NT levels were significantly lower (p<.05, p<.05 and p<.001, respectively), whereas serum and aqueous humor DS levels were significantly higher in cataract patients (p<.01 and p<.001). DS/TT and DS/NT ratios were significantly higher and the NT/TT ratio was lower in cataract patients in serum (p<.005) and aqueous humor samples (p<.001). In conclusion, serum T-D homeostasis may be useful as biochemical markers, indicating the role of oxidative stress in the development of cataracts. Methods: During the 2014-2015 academic school year, 381 physiotherapy students were enlisted from two universities in Istanbul, one a foundation and the other a government university. The Turkish version of the Jefferson Scale of Physician Empathy was administered. Students were asked to indicate interest in particular physiotherapy specialties as well as their region of origin in Turkey. The Kruskal-Wallis analysis was used to determine differences among the four study years, and also to measure relationships between specialty interest, home-region, and empathy scores of the students. Empathy scores were also compared according to gender. Results: The difference of empathy scores between the students of the two universities was borderline significant (p=0.057). Empathy scores in both universities increased to a significant degree after school entrance and decreased in the final year. Levels of empathy did not change according to gender, specialty interest, or home-region (p=0.722, 0.524, and 0.309, respectively). Conclusions: This study points to the need for physiotherapy curricula that would enhance empathy and give students practice in exhibiting this valuable attribute. Additional studies are needed that would include larger study populations and track the same students year by year as to how and why their empathy levels change during their training.Publication A skeletochronological study of age, growth and longevity in two freshwater turtles, Emys orbicularis and Mauremys rivulata from Mediterranean Turkey (Reptilia: Testudines).(2016-02-01) Ayaz, Dinçer; Çiçek, Kerim; KUMAŞ, MELTEM; Tok, Cemal Varol; KUMAŞ, MELTEMWe estimated growth parameters of Emys orbicularis and Mauremys rivulata in Mediterranean Turkey with the skeletochronological method, using specimens drowned in fyke nets. In adult E. orbicularis, the median age was 8 years in males and 10 years in females. The median age of adult M. rivulata was 10 years for both sexes. Both species reach sexual maturity at an age of 5-7 years. To assess the prognostic value of age on the outcome of transcanalicular multidiode laser dacryocystorhinostomy (TCL-DCR) in patients with acquired nasolacrimal duct obstruction (NLDO). Methods. The medical records of TCL-DCR performed between March 2009 and September 2013 were reviewed retrospectively. Inclusion criteria include over 20 years of age, similar mean follow-up period, and similar mean duration of stenting. The main outcome is surgical success. The effect of age on success rate is also evaluated. Results. The anatomical success was 52% in Group 1 (20-30 years), 56% in Group 2 (31-40 years), 64% in Group 3 (41-50 years), 76% in Group 4 (51-60 years), and 88% in Group 5 (over 60 years). The statistical difference among Group 1 and Group 5, in terms of surgical success rate, was found to be significant (P = 0.009). Additionally, the 20-30-year-old patients had a failure rate 6.76 times higher than that of the over-60-year-old patients (P = 0.009; 95% CI, 1.605-28.542). Conclusion. TCL-DCR is a surgical treatment option for NLDO for which a skin incision can be avoided. However, the mechanism(s) of this effect has not been elucidated yet. In this study, cytotoxic, genotoxic, apoptotic and reactive oxygen generating (ROS) activities of ABS were investigated in melanoma and normal cell lines. The cells were incubated with different concentrations of ABS (0.125 to 2%) for 24 h. The cell viability was assessed based on ATP cell viability assay. Intracellular accumulation of reactive oxygen species (ROS) was determined using the fluorescent probes 2-,7--dichloro-dihydrofluoresce-in-diacetate (H2DCF-DA). DNA damage was evaluated by alkaline single cell gel electrophoresis assay (Comet Assay) and, apoptosis induction was detected by Acridine Orange/Ethidium Bromide AO/EB double staining method. Our results demonstrated that ABS increases DNA damage, apoptosis and ROS levels in both melanoma and normal cell lines in a dose dependent manner, and all of these activities were significantly higher in melanoma cells than in normal cells. There was a statistically significant positive correlation between DNA damage, apoptosis and ROS levels in ABS treated cell lines. Our results revealed that although ABS commonly used as hemostatic agent, it causes DNA damage and apoptosis by generating ROS in a dose dependent manner. Therefore, it should be removed the unused ABS by cleaning once the hemostasis is achieved to minimize the postoperative side effects. These results could also contribute to the development of new treatment for cancer.Publication Anti-inflammatory and antioxidative properties of honey bee venom on Freund’xxs Complete Adjuvant-induced arthritis model in rats(2019-04-01) KOÇYİĞİT, ABDÜRRAHİM; GÜLER, ERAY METİN; KALELİ, SÜLEYMAN; KOÇYİĞİT, ABDÜRRAHİM; GÜLER, ERAY METİNThe aim of this study was to assess anti-inflammatory, anti-oxidant, anti-genotoxic and immunomodulatory effects of honey bee venom (HBV) on adjuvant-induced arthritis in rats. Thirty-five rats were equally divided into a negative control (NC), a positive control (PC) and low, moderate and high doses (2, 4 and 20 mg/kg, respectively) of HBV treatment groups. Freund-s Complete Adjuvant (FCA) was given to the rats to form arthritis. The treatment groups were treated with HBV for 3 consecutive weeks. After the treatment, plasma IL-1 beta, IL-6, TNF-alpha, IFN-gamma and TGF-beta 1, total oxidant status (TOS), total antioxidant status (TAS) and myeloperoxidase (MPO) activities and mononuclear leukocyte (MNL) DNA damage levels were measured. Oxidative stress index (OSI) was calculated. IL-beta, IL-6, TNF-alpha, TGF-beta 1, IFN-gamma, TOS, OSI, DNA damage levels and MPO activities were significantly higher and TAS levels were lower in the PC group than the NC. After low-doses of HBV treatment IL-1 beta, IL-6, TNF-alpha, TGF-beta 1, TOS, OSI, MPO and MNL-DNA damage levels significantly decreased according to the PC, while IFN-gamma and TAS levels increased. The differences in moderate and high-dose HBV treatment groups were not as significant as low HBV doses. Low-doses of HBV has been shown to treat RA with anti-inflammatory and antioxidant effects, by preventing DNA damage. However, these effects have not been observed as strong at higher doses of HBV.
