Abstract
Nowadays, there are many wound healing therapy methods but yet to increase rate of wound contraction, to provide least scar forming and to decrease feeling pain lots of studies on wound and wound healing are going on. In recent years, it is revealed that the ingredients of PRP have an intensifier effect on regeneration and therefore PRP treatment has become a popular topic for wound healing. Protein and lipid structured bioactive biomolecules are components of PRP and they accelerate healing via stimulating wound healing at the molecular level. According to an in vitro study in literature, during wound healing protein structured components of PRP are being degraded by protease enzymes' activity, therefore healing effect of PRP is going on with lipid structured components of PRP that is lipid portion of PRP. On the basis of this hypothesis, thesis's idea is designated as to evaluate the macroscopic and molecular effects of lipid portion of PRP on the wound healing via in vivo experimental set up. For macroscopic testing, wound contraction calculations dependent on nwound areas were registered while for the molecular testing, mRNA expressions of 84 different genes related with wound healing were analyzed. mRNA that is RNA molecule trasporting genetic code for protein synthesis, assesment of mRNA expression profiles provide the information about the molecular level protein synthesis during the wound healing.Animal experiments part of our study have been completed with 20 Sprague Dawley rats as four different groups with 1.2 diameter circular full thickness wounds were defected on their's dorsum region. Groups were PRP, lipid portion, DMSO and SHAM (as control).Then, biopsy samples have been taken from wounds on 3., 7., ve 14. days. After mRNA isolation from biopsy samples and cDNA synthesis from mRNA, w a PCR array technique, real-time PCR, were made to analyse expression levels of genes related with wound healing. 2ˆΔΔCT procedure has been used with fold change values to determine which genes upregulate and downregulate their expression. When regulations' results were analyzed with respect days of biopsy taken, the most number of gene expression alterations were seen at proliferation phase as percentaged. Although PRP and lipid portion treatment groups show similar regulation at the stages of wound healing as inflamation, proliferation and maturation, PRP group's fold changes on some genes were more than lipid portion. According to macroscopic evaluations, rate of wound contraction of lipid portion treatment group's wounds were less than PRP treatment group's wounds. PRP group's wound contraction data were analyzed with T-Test and it was meaningful statistically. As a result, RT-PCR data belong to PRP and lipid portion treatment groups have appeared similar at the molecular level, lipid portion obtained from PRP has component and healing effect of PRP but PRP is more effective on the wound healing.
