dc.description.abstractVasospasm in free tissue transfers is a serious problem that can cause necrosis, which impairs flap perfusion.A lot of surgical techniques and medications were used to resolve vasospasm. In this study, our aim is to evaluate the flap perfusion by applying magnesium sulfate both into intravenous and intravenous 40 sprague dawley rats were divided into 4 groups.Groin flap was planned for all experimental rats.Vasospasm has been carried out by pedicle traction. Femoral artery transection and anastomosis are performed. Normal saline was applied on the vessel of group 1, 10% magnesium sulphate in the 2nd group, Intraarterial 10% magnesium sulphate in the 3nd group, 10% magnesium sulphate on vessel and intraarterial in the 4th group respectively. Tissue perfusion was measured by laser doppler flowmeter (LDF) before, during, and following vasospasm before sacrification on 7 day.. The degrees of EGF, VEGF, MMP-2, MMP-9, HIF-1 alpha, cyclophilin-A, TAS and TOS were monitored in samples of tissue. Following anastomosis, patency was evaluated on 7th.day In group 4, flap perfusion and angiogenesis markers were found to be significantly higher, while ischemia-reperfusion injury and apoptosis were found to be significantly lower (p˂0,005). There was no difference in terms of anastomotic patency among the groups. In this study,with the application of Magnesium sulfate bothinto intra-arterial and onto the anastomosis, it was observed that vasospasm improved more effectively compared to others.According to these results,it was concluded that this treatment model can us this treatment model can be used for clinical and experimental free tissue transfer studies.en
