Abstract
Aside from its pervasiveness, whereby it affects as much as 20% of the world's population, depression continues to be one of the most important psychiatric problems due to the loss of power caused by disrupting daily life functioning, economic consequences, and high suicidal tendency. A diagnostic system based on biochemical techniques for major depression has not yet been developed. In this study, we aimed to identify the proteins in serum samples that change during depression with the major depression model created by forced swimming test in rats. Serum samples taken from 14 animals, as divided by depression and control groups, were separated by chromatographic methods and then compared with 2D electrophoresis. After the two-dimensional electrophoresis, a total of 9 potential diagnostic protein sequences were identified, which differed in their expressions with computer software. During the last phase of the study of the MALDI-TOF analysis, the sequences of points identified differentially among the groups were determined by calculating the durations of amino acid residues in the electrical field. By comparing protein expressions with significant differences among them, it was concluded that 9 different points could be used together as a potential biomarker.

Depresyon, dünya nüfusunun yaklaşık %20 gibi büyük bir kısmını etkileyecek kadar yaygın olması yanında; günlük yaşam işlevselliğini bozarak neden olduğu yeti kayıpları, ekonomik sonuçları ve yüksek intihar eğilimi nedeniyle de en önemli psikiyatrik sorunlardan biri olma özelliğini sürdürmektedir. Major depresyon tanısında biyokimyasal tekniklere dayalı bir tanı sistemi henüz geliştirilmemiştir. Bu çalışmanın amacı, sıçanlarda zorunlu yüzme testi ile oluşturulmuş depresyon modeli ile depresyon sırasında değişim gösteren serum örneklerindeki proteinlerin tanımlanmasıdır. Depresyon ve kontrol gurubu olarak ayrılan toplam 14 hayvandan alınan serum örnekleri kromatografik yöntemlerle ayrıştırılıp daha sonra 2 boyutlu elektroforez ile karşılaştırılması yapıldı. İki boyutlu elektroforez sonrasında bilgisayar yazılımı ile ifadelerinde farklılık gösteren toplam 9 potansiyel tanı proteini dizisi belirlendi. Çalışmanın son aşaması olan MALDI-TOF analizi aşamasında, guruplar arasında farklı ifade edildiği belirlenen noktaların dizileri elektriksel alanda aminoasit kalım sürelerini hesaplama yoluyla belirlendi. Aralarında anlamlı farklılık bulunan protein ifadeleri karşılaştırılarak elde edilen 9 farklı noktanın ayrı ayrı ya da birkaçı birlikte kullanılarak potansiyel biyobelirteçler olabilecekleri sonucuna varıldı
