    The Nutritional Habits and Relationship Between The Antioxidant Activity and Iron Deficiency Anemia During Pregnancy

All rights reserved.The aim of this study was to investigate the changes in nutritional habits, and also relationship among oxidative stress, iron deficiency anemia and some antioxidant enzyme activities during pregnancy. Twenty pregnant women in the first trimester, 20 pregnant women in the last trimester and 20 non-pregnant women (control group) were included in the study. The study groups were similar in terms of age and body mass index. None of the participants had any systemic disease or smoked. After having filled out the questionnaire about nutritional habits, the blood samples were collected, and the malondialdehyde (MDA) level and activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) enzymes were measured together with hemoglobin (Hgb), hematocrit (Hct), iron, ferritin and the mean erythrocyte volume (MCV) levels. The most frequent meat consumption was on every other day in controls, but once a week in the first and last trimester groups. In addition, the most frequent egg consumption was on every other day and once a week in contro ls, but rare in the first trimester group, and once a week in the last trimester group. Although iron and vitamin use was 10% and 30% in the first trimester group, it was 50% and 65% respectively in the last trimester group. The MDA level was higher, and t he SOD, CAT and GSH-Px enzyme activities were lower in both pregnant groups than controls. The Hgb and Hct values were lower in the last trimester group compared to the controls and the first trimester group. The first trimester group and the controls were similar in terms of Hgb and Hct levels. There was no significant difference among groups in terms of ferritin, iron or MCV values. It was concluded that; a) the egg and meat consumption might decrease with the progress of pregnancy, b) the oxidative stress increased in pregnancy, c) Hgb and Hct values might be lower in the last trimester group despite increased intake of oral iron and vitamin supplements, and normal serum iron levels due to decreased CAT activity. Therefore, more studies are needed to demonstrate the relationship among oxidative stress, anemia and CAT enzyme activity in pregnancy.

    Pentylenetetrazole-induced kindling rat model: miR-182 and miR-27b-3p mediated neuroprotective effect of thymoquinone in the hippocampus

    Objectives: Epilepsy is a neurological disease that pathologically affects brain functions. The epileptic hippocampus has modified microRNA(miRNA) levels. Therefore, we aimed to evaluate the neuroprotective effect of thymoquinone (TQ) in PTZ-induced epilepsy and to demonstrate the overlap between miRNA and mRNA expression profiles. Methods: Male adult Wistar albino rats (200-230 g, n = 20) were divided into three groups as control (n = 6), PTZ (n = 7), and TQ + PTZ (n = 7). The PTZ kindling model was created by injecting PTZ in sub convulsive doses to rats on days 1, 3, 5, 8, 10, 12, 15, 17, 19, 22, and 24 of the study into animals. Clonic and tonic seizures were induced by injecting a convulsive dose of PTZ on day 26 of the study. Rats in the TQ+PTZ group were treated by oral gavage with a 20 mg/kg TQ 2 h before each PTZ injection. The rats in the control group were treated with 0.5 ml saline. Seizure severity was evaluated with the Racine scale. The genes and signaling pathways targeted by miRNAs were determined by bioinformatics analysis. Results: In the rat hippocampus, mature 728 miRNAs were analyzed by microarray and the nine miRNA were verified by quantitative Real-Time PCR. rno-miR-182 and rno-miR-27b-3p were up-regulated in the PTZ group and down-regulated in the TQ + PTZ group. Discussion: In the PTZ kindling epilepsy model, the expression of these two miRNAs was regulated by TQ and exerted a neuroprotective effect by controlling the activities of target genes.

    Angiotensin IV improves spatial memory in streptozotocin-induced diabetic rats by reducing oxidative stress and altering BDNF levels

    In this study, we investigated the protective effects of angiotensin IV (Mg IV) on cognitive function in streptozotocin (STZ)-induced diabetic rats. Male Wistar albino rats, were randomly divided into four groups; control (C), diabetes (Dia, 60 mg/kg, STZ, i.p.), Mg IV (5 mu g/kg, s.c.) and Dia+Ang W. The passive avoidance and Morris water maze (MWM) tests were used to evaluate learning and memory performance. Behavioral tests were carried out between 21 and 30 days after the initial Ang IV injection. Hippocampi were dissected and retained for biochemical and Western blot analysis. The Dia group exhibited the poorest behavioral results, while the Dia+Ang W group performed highest on the MWM task. Superoxide dismutase, glutathione peroxidase, and malondialdehyde levels increased significantly in the Dia group compared to Dia+Ang IV. Brain-derived neurotrophic factor (BDNF) and N-methyl-D-aspartate levels were significantly elevated, while levels of GABA(A) significantly decreased, in the Dia+Ang IV group compared to the Dia group. These findings suggest that peripheral administration of Ang IV ameliorated spatial memory in diabetic rats by decreasing hippocampal oxidative stress and BDNF levels.

    Impaired memory by hippocampal oxidative stress in rats exposed to 900 MHz electromagnetic fields is ameliorated by thymoquinone

    The protective effects of thymoquinone on learning and memory abilities and on hippocampal oxidative stress in rats exposed to 900 MHz electromagnetic field radiation at s strength of 6.0 +/- 0.5 V/m for 1 h on each of 15 days was studied. Thymoquinone-treatment (10 mg/kg/day) of radiation-exposed rats resulted in better performance than that of radiation-exposed rats not receiving thymoquinone. Electromagnetic field-exposure led to increased hippocampal superoxide dismutase, glutathione peroxidase, and malondialdehyde levels and to lowered brain-derived neurotrophic factor level; thymoquinone treatment ameliorated all four parameters. Also, in the electromagnetic field-exposed group, N-methyl-D-aspartate receptor expression decreased, and gamma-aminobutyric-acid alpha receptor expression increased, which was reversed by thymoquinone treatment. In conclusion, treatment with the thymoquinone ameliorated the impact of the exposure to EMF and provided a hint on the mechanism involving oxidative stress.

    Effects of Thymoquinone On Erythrocyte Aggregation Kinetics: An In Vitro Study

All rights reserved.Erythrocyte aggregation is associated with a series of physio-pathological processes in body and according to this, current study was designed to examine how the thymoquinone (TQ) and a ggregation kinetics relation changes when the aggregation properties of erythrocyte suspensions are intervened using different approaches, such as changing the suspension environment or erythrocyte surface properties. Blood samples from 40 male Wistar albino rats (three months old weighing 250-300 g) were randomly divided into four groups, as 1) whole blood (control), 2) 1/2 dilution (plasma was diluted with PBS), 3) dextran 500 (1% dextran 500 was dissolved in plasma) and 4) glutaraldehyde. Blood samples were taken from abdominal aorta under anesthesia. The erythrocyte suspensions were incubated at 37°C for one hour in the absence or presence of TQ, with a final concentration of 1 mg/ml. Then, the electrical properties of erythrocyte suspensions were recorded, and the electrical capacitance was monitored using an inductance, capacitance, and resistance meter (LCR meter) at 100 kHz. It was found that TQ incubation decreased the aggregation index in control and dextran groups while increased it in 1/2 dilution and GA groups. It was also found that TQ incubation increased the time constant in whole blood and dextran while decreased it in 1/2 dilution and GA. The TQ can normalize the altered erythrocyte aggregation responses in both ways that is, it decreases the increased aggregation or increases the decreased aggregation. Therefore, it may have very critical role in rouleaux formation-induced circulatory disorders.

    A low direct electrical signal attenuates oxidative stress and inflammation in septic rats

    Electrical stimulation is proposed to exert an antimicrobial effect according to studies performed using bacterial and cell cultures. Therefore, we investigated the effects of electrification on inflammation in septic rats. Twenty-eight male Wistar albino rats were divided into 4 groups: healthy control (C), electrified healthy (E), sepsis (S), and electrified sepsis (SE) groups. Staphylococcus aureus (1 x 109 colonies) in 1 ml of medium was intraperitoneally injected into rats to produce a sepsis model. The rats in the E and SE groups were exposed to a low direct electrical signal (300 Hz and 2.5 volts) for 40 min and 1 and 6 h after bacterial infection. Immediately after the second electrical signal application, blood and tissue samples of the heart, lung, and liver were collected. An antibacterial effect of a low direct electrical signal was observed in the blood of rats. The effects of electrical signals on ameliorating changes in the histological structure of tissues, blood pH, gases, viscosity and cell count, activities of some important enzymes, oxidative stress parameters, inflammation and tissue apoptosis were observed in the SE group compared to the S group. Low direct electrical signal application exerts antibacterial, antioxidant, anti-inflammatory and antiapoptotic effects on septic rats due to the induction of electrolysis in body fluids without producing any tissue damage.
