Ten 12-week-old Wistar albino rats were used for this preliminary study. MSCs were obtained from bones of two animals and were labeled with green fluorescent protein (GFP). Four animals were randomly selected for MSC injection, while 4 animals served as a control group. Samples were prepared for histological analysis, Cox-2 mRNA expression polymerase chain reaction analysis, and fluorescent microscopy evaluation. The number of total cells, number of osteoclastic cells, and Cox-2 mRNA expression levels of the periodontal tissue of teeth were calculated. The number of total cells was increased with MSC injections in PDL significantly (P < 0.001). The number of osteoclastic cells and Cox-2 mRNA expression were found to be similar for the two groups. GFP-labeled MSCs were observed with an expected luminescence on the smear samples of the PDL with transferred MSCs. Seventy decoronated single-rooted human teeth were infected and divided into 6 test (n=10) and 2 control groups (n=5) (negative, sterile samples and positive, infected samples). Following irrigants were then applied to test groups: 2.5% NaOCl, 5.25% NaOCl, CHX, CHX+CTR, S. officinalis extract and OCT. The dentin chips were obtained from inner root canal walls and analyzed by counting the number of colony forming units (CFU). The 2.5% NaOCl, 5.25% NaOCl, CHX and OCT groups presented no bacterial growth (CFU=0). S. officinalis and CHX+CTR groups reduced the number of E. faecalis cells but could not eliminate all. Single-rooted teeth were decoronated and restored with ParaPost (PP), i-TFC (TFC), and DPs (n=10). Resin cores were created and loaded until failure (1.0mm/min). The data were analyzed (ANOVA, Tukey, Chi-Square tests). Three-dimensional FEA models of the posts were created and the stress distributions were calculated using Solid Works/Cosmos works structural analysis program. A significant difference was found among the groups (p<0.001). TFC group showed the highest and the DP group showed the lowest fracture resistance (p<0.001). PP created more stress at the root dentin and inside the post. Restoration of roots with glass fiber and DPs may reduce stresses within the core material and the remaining root. Araştırmacıların başvuru dosyasındaki belgeleri eksiksiz tamamlamadaki isteksizliği, Etik Kurulların çalışma sürecini yavaşlatmakta ve hatta Etik Kurul ile araştırmacılar arasında tartışmaya bile neden olabilmektedir. Bu nedenle bu çalışma, bir üniversitenin Girişimsel Olmayan Araştırmalar Etik Kuruluna gönderilen proje başvuru dosyalarındaki eksiklikleri ve en çok yapılan hataları göstermek ve araştırmacılar için bir rehber oluşturmak amacıyla yapıldı. 24 Mart 2016 ve 30 Haziran 2018 tarihleri arasında Girişimsel Olmayan Araştırmalar Etik Kurula gönderilen başvuru dosyaları ve kararları ilgili etik kurulun veri tabanından analiz edildi. Başvuru dosyalarının revizyonuna neden olan en önemli gerekçeler, projenin amacının ilgili referanslar eşliğinde yeterince açıklanamaması ve özellikle iyi yazılamamış yöntem kısmı gibi başvuru formundaki eksikliklerdi. Materials and methods: Eighty human mandibular premolar teeth were randomly divided into eight groups: Group IN, Group BF, Group PRF1, Group PRF2, Group IN-TMA, Group BF-TMA, Group PRF1-TMA ,and Group PRF2-TMA. Group IN (intact) and Group IN-TMA (intact but subjected to thermomechanical aging) served as control groups. In the other six groups, endodontic treatment was performed and standardized mesio-occluso-distal (MOD) cavities were prepared. In BF, PRF1, and PRF2, the cavities were restored with bulk-fill composite only, bulk-fill/Ribbond, and bulk-fill/additional Ribbond, respectively. In BF-TMA, PRF1-TMA, and PRF2-TMA, the teeth were subjected to thermomechanical aging after the restorations. All of the teeth were fractured on the universal testing machine. Fracture surfaces were analyzed with a stereomicroscope. Results: Control groups showed significantly higher fracture strengths than tested groups (P<0.05). No statistically significant difference was observed among the tested groups (P>0.05). Most of the favorable fractures were seen in PRF1, PRF2, and PRF2-TMA. Most of the unfavorable fractures were seen in BF-TMA. Conclusions: Although fiber insertion with different techniques did not increase the fracture strength of teeth restored with bulk-fill composites, it increased the favorable fracture modes.
