However, few studies have addressed its role in odontoblastic differentiation of human dental pulp-stem cells (hDPSCs). Aim: This study aimed to understand the influence of various concentrations of 1,25-OH D-3 on the proliferation capacity and early dentinogenesis responses of hDPSCs. Materials and Methods: hDPSCs were obtained from the impacted third molar teeth. Monolayer cultured cells were incubated with a differentiation medium containing different concentrations of 1,25-OH D-3 (0.001, 0.01, and 0.1 mu M). All groups were evaluated by S-phase rate [immunohistochemical (IHC) bromodeoxyuridine (BrdU) staining], STRO-1 and dentin sialoprotein (DSP)+ levels (IHC), and alkaline phosphatase (ALP, enzyme-linked immunosorbent assay (ELISA)) levels. Results: The number of cells that entered the S-phase was determined to be the highest and lowest in the control and 0.001 mu M 1,25-OH D-3 groups, respectively. The 0.1 mu M vitamin D-3 group had the highest increase in DSP+ levels. The highest Stro-1 levels were detected in the control and 0.1 mu M 1,25-OH D-3 groups, respectively. The 0.1 mu M 1,25-OH D-3 induced a mild increase in ALP activity. Conclusions: This study demonstrated that 1,25-OH D-3 stimulated odontoblastic differentiation of hDPSCs in vitro in a dose-dependent manner. The high DSP + cell number and a mild increase in ALP activity suggest that DPSCs treated with 0.1 M 1,25-OH D-3 are in the later stage of odontoblastic differentiation. The results confirm that 1,25-OH D-3-added cocktail medium provides a sufficient microenvironment for the odontoblastic differentiation of hDPSCs in vitro.

METHODS Two experienced surgeons performed 120-W HPS laser PVP. The baseline characteristics, perioperative data, complications, and postoperative outcomes were evaluated at 3, 6, 12, 24, and 36 months postoperatively. RESULTS A total of 550 consecutive patients, with a mean age of 67.6 years, underwent PVP with a 120-W HPS laser. The mean prostate volume was 72.93 cm(3), with a mean prostate-specific antigen level of 3.57 ng/mL. The mean operative duration and the mean applied energy was 61.3 minutes and 164.06 kJ, respectively. No major complication occurred intraoperatively or postoperatively. The mean follow-up was 17.80 months. Significant improvements were observed postoperatively in the mean International Prostate Symptom Score, quality of life score, maximal urinary flow rate, and postvoid residual urine volume. The mean catheterization time was 18.5 hours. The complications included delayed hematuria in 26 (4.8%), recatheterization in 24 (4.4%), reoperation owing to residual prostatic adenoma in 46 (8.5%), urethral stricture in 19 (3.5%), and bladder neck contracture in 6 (1.1%) patients. CONCLUSIONS PVP with a 120-W HPS laser for benign prostatic hyperplasia has been proved to be a safe and effective procedure for our patients, including those treated with oral anticoagulants. - PublicationMetadata only16-kDa Prolactin Promotes Cardiac Ischemia/Reperfusion Injury(22.10.2013) Yamac, Aylin HaticeThe recent discovery that anti-angiogenic, pro-apoptotic and pro-inflammatory N-terminal 16-kDa prolactin subform (16-kDa Prl) is generated under enhanced oxidative stress and yielding adverse cardiac effects, prompted us to investigate the potential role of prolactin and its cleaved 16-kDa form in myocardial ischemia/reperfusion (I/R) injury. In the current study we demonstrate that enhanced levels of serum prolactin (Prl) and cathepsin D (CD) activity in patients with acute myocardial infarction (AMI) were associated with the generation of N-terminal 16-kDa subform. In a murine model of myocardial I/R with previous LAD (left anterior descending artery) ligation for 50 minutes, blockage of endogenous Prl release by the dopamine D2 receptor agonist bromocriptine limited infarct size 24 hours post-reperfusion and preserved left ventricular (LV) function after 14 days of reperfusion. Additionally, we demonstrate that after blockage of endogenous Prl release, the subsequent application of a recombinant mutant, prolactin isoform, which is not cleaved into the 16-kDa subform, significantly limited the extent of cardiac injury compared to the well-cleaved corresponding wildtype prolactin isoform 24 hours post-reperfusion. I/R injury-induced up-regulation of pro-inflammatory mediators (e.g. TNF-α, CCL-2, CXCL2) and extravasation of inflammatory infiltrates were significantly reduced in bromocriptine treated mice than untreated controls. The bromocriptine induced cardiac protection was mainly addicted to inhibition of 16-kDa Prl formation, as myocardium treated with the mutant prolactin isoform displayed substantially less inflammatory cell infiltration and cytokine expression than myocardium treated with the wildtype prolactin isoform. In addition, we discovered that recombinant 16-kDa Prl markedly induced the expression of proinflammatory cytokines via activation of NF-κB signalling in neonatal rat cardiomyocytes.

Yunus; Uckaya, Fatih; Topcu, Gulacti; Esrefoglu, Mukaddes; Bayindir, NihanA novel automatic Rapid Diagnostic Test (RDT) reader platform is designed to analyze and diagnose target disease by using existing consumer cameras of a laptop-computer or a tablet. The RDT reader is useable with numerous lateral immunochromatographic assays and similar biomedical tests. The system has two different components, which are 3D-printed, low-cost, tiny, and compact stand and a decision program named RDT-AutoReader 2.0. The program takes the image of RDT, crops the region of interest (ROI), and extracts the features from the control end test lines to classify the results as invalid, positive, or negative. All related patient's personal information, image of ROI, and the e-report are digitally saved and transferred to the related clinician. Condition of the patient and the progress of the disease can be monitored by using the saved data. The reader platform has been tested by taking image from used cassette RDTs of rotavirus (RtV)/adenovirus (AdV) and lateral flow strip RDTs of Helicobacter pylori (H. pylori) before discarding them. The created RDT reader can also supply real-time statistics of various illnesses by using databases and Internet. This can help to inhibit propagation of contagious diseases and to increase readiness against epidemic diseases worldwide.

VOCAL programme, echogenicity, number of follicles and blood flow parameters were evaluated. The patients with PCOS had a higher total ovarian volume, mean stromal volume and stromal echogenicity (18.6 +/- 4.75 to 10.2 +/- 3.4p .05). 3 D power Doppler parameters included VI, FI, and VFI values of the patients with PCOS were higher when compared to those of the patients with multifollicular ovary (3.82 +/- 2.65 to 1.78 +/- 1.2,p < .01; 50.76 +/- 4.45 to 40.6 +/- 3.64,p = .03; and 2.34 +/- 1.02 to 1.12 +/- 0.65,p = .02, respectively). Our results revealed that total ovarian volume, stromal volume and echogenicity; VFI, VI, and FI could be useful for differential diagnosis in women with PCOS and multifollicular ovaries.Impact statement What is already known on this subject?Ultrasonography is considered the new diagnostic tool for PCOS. Enlarged ovaries with multiple small follicles peripherally located around increased ovarian stroma with increased stromal echogenicity are the sonographic features of polycystic ovaries. What do the results of this study add?3-D Doppler ultrasonography may be more specific in the determination of multifollicular and polycystic ovaries when compared to RI and PI in 2-D Doppler ultrasonography. Moreover, 3-D power Doppler ultrasonography could be useful for differential diagnosis in women with PCOS and multifollicular ovaries. What are the implications of these findings for clinical practice and/or further research?In clinical practice, the differentiation of multifollicular ovaries and polycystic ovaries (PCO) is difficult with the use of 2-D sonography alone. Therefore, 3-D ultrasound and power Doppler may also be used in addition to 2-D ultrasound for the differentiation of multifollicular ovaries and PCO.
