- PublicationMetadata onlySpectrofluorimetric Methods for the Determination of Gemifloxacin in Tablets and Spiked Plasma Samples(2011-05-01) Tekkeli, Serife Evrim Kepekci; Onal, Armagan; TEKKELİ, ŞERİFE EVRİMTwo new, sensitive and selective spectrofluorimetric methods have been developed for the determination of gemifloxacin (GFX) in tablets and spiked plasma samples. Gemifloxacin, as a primary amine compound, reacts with 7-chloro-4-nitrobenzofurazon (NBD-Cl) (for method A) and fluorescamine (for method B) which are a highly sensitive fluorogenic reagents used in many investigations. For method A, the reaction product was measured spectrofluorimetrically at 516 nm with excitation at 451 nm. The reaction proceeded quantitatively at pH 8.5, 80 A degrees C in 7 min. For method B, the method was based on the reaction between GFX and fluorescamine in borate buffer solution of pH 8.5 to give highly fluorescent derivatives that were measured at 481 nm using an excitation wavelength of 351 nm. The fluorescence intensity was directly proportional to the concentration over the range 40-200 ng mL(-1) and 100-1,200 ng mL(-1) for method A and B, respectively. Successful applications of the developed methods, for the drug determination in pharmaceutical preparations and spiked plasma samples, were performed.

- PublicationMetadata onlyEnt-kaurane diterpenoids isolated from Sideritis congesta(2011-12-01) Topcu, GÜLAÇTI; Ertas, Abdulselam; Ozturk, Mehmet; Dincel, Demet; Kilic, Turgut; Halfon, Belkis; TOPÇU, GÜLAÇTI; DİNCEL, DEMETA new ent-kaurane diterpenoid, together with eight known ent-kauranes, were isolated from the petroleum ether and acetone extracts of the whole plant of Sideritis congesta P. H. Davis & Hub.-Mor. and their structures were elucidated as the new compound ent-7 alpha-acetoxy-16 beta, 18-dihydroxy-kaurane (7-acetyldistanol) (1) and the known compounds ent-3 beta,7 alpha-dihydroxy, 18-acetoxy-15 beta,16 beta-epoxykaurane (epoxyisolinearol) (2), sideroxol (3), sideridiol (4), siderol (5), 7-epicandicandiol (6), foliol (7), linearol (8) and sidol (9). Characterization of compounds 1-9 was based on spectral analyses and comparison with reported data, particularly the new compound 1 was identified by 1D- and 2D-NMR and mass spectroscopic analyses. The antioxidant potential of the extracts, and also of the ent-kauranes except for 7, was investigated by three methods including beta-carotene bleaching method, free radical scavenging activity and superoxide anion scavenging activity. The anticholinesterase activity was also evaluated for the ent-kauranes except for 7, and most of the diterpenes exhibited weak acetylcholinesterase inhibitory activity. However, almost all diterpenes exhibited some inhibitory activity against butyrylcholinesterase; particularly, compounds 3 and 6 exhibited better BChE inhibitory activity than the standard compound galanthamine. H. Davis have been studied for their diterpenic compounds and the antioxidant properties. Eight known diterpenoids, which have ent-kaurene skeleton, were isolated from acetone and methanol extracts of these species. The structures of the isolated diterpenes were determined by using the NMR (H-1-NMR, C-13-NMR, COSY, HMQC, and HMBC) spectroscopy. The analysis of the phenolic compounds of the extracts was performed by high performance liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). Also the antioxidant capacity of the extracts was investigated namely by two methods; free radical scavenging and beta-carotene bleaching activities.

- PublicationMetadata onlyA Review of Current Methods for the Determination of Acrylamide in Food Products(2012-02-01) Tekkeli, Serife Evrim Kepekci; Onal, Cem; Onal, Armagan; TEKKELİ, ŞERİFE EVRİMAcrylamide (AA) is a potentially carcinogenic substance which is formed during heating of food products containing carbohydrates and asparagine. It was first detected in food products in 2002. Since that time, several analytical methods have been made available for the quantification of AA in various foods. Starting from the announcement in 2002, occurrence, formation, chemistry, toxicology, and potential health risk in the human diet have been investigated and methods of analysis have been reviewed in many articles. In this paper, current information and analytical methods for the determination of AA have been reviewed.
- PublicationMetadata onlyMorphological, anatomical and ecological studies on some Orchis (Orchidaceae) taxa of Mediterranean region, Turkey(2012-04-01) Sevgi, ECE; Altundag, Ernaz; Kara, Omer; Sevgi, Orhan; Tecimen, Huseyin Baris; Bolat, Ilyas; SEVGİ, ECEIn this study, nine species mainly distributed in Mediterranean (+ Aegean region) region of Turkey were investigated in terms of 15 morphological, 16 anatomical and 5 ecological characteristics. Those species are Orchis anatolica Boiss., Orchis italica Poiret, Orchis laxiflora Lam., Orchis mono L. subsp. mono, Orchis provincialis Balbis ex DC., Orchis purpurea Hudson, Orchis sancta L., Orchis simia Lam., Orchis tridentata Scop. In conclusion, we found that O. laxiflora was characterized by the longest plant height and O. purpurea had the biggest tuber; whereas O. italica was identified by the highest number of leafs and O. purpurea had the longest leaf length and widest leaf width. Additionally, the other species which were characterized by different morphological and anatomical parameters are as follow: O. sancta with the longest bract length, O. italica and O. simia with the longest sepal lengths, O. laxiflora with the shortest and widest labellum, O. simia with the longest petal length, O. sancta with longest caudiculum length, O. anatolica with the longest spur length and O. provincialis with the longest ovary length. Particularly, O. laxiflora and a purpurea species present essential divergence from the aspect of anatomical features of leaf surface in comparison with the other species. Morphological and anatomical traits of the species were attributed to the habitat selections of the species since that character differs along with each species.
